Abstract: Current methods determining biomass yield require sophisticated sensors for in situ measurements or multiple steady-state reactor runs. Determining the yield of specific groups of organisms in mixed cultures in a fast and easy manner remains challenging. This study describes a fast method to estimate the maximum biomass yield (Ymax), based on 13C incorporation during activity measurements. It was applied to mixed cultures containing ammonia oxidizing bacteria (AOB) or archaea (AOA) and nitrite oxidizing bacteria (NOB), grown under mesophilic (1528∘C) and thermophilic (50∘C) conditions. Using this method, no distinction could be made between AOB and AOA co-existing in a community. A slight overestimation of the nitrifier biomass due to 13C redirection via SMP to heterotrophs could occur, meaning that this method determines the carbon fixation activity of the autotrophic microorganisms rather than the actual nitrifier biomass yield. Thermophilic AOA yields exceeded mesophilic AOB yields (0.22 vs. 0.060.11 g VSS g-1 N), possibly linked to a more efficient pathway for CO2 incorporation. NOB thermophilically produced less biomass (0.0250.028 vs. 0.0480.051 g VSS g-1 N), conceivably attributed to higher maintenance requirement, rendering less energy available for biomass synthesis. Interestingly, thermophilic nitrification yield was higher than its mesophilic counterpart, due to the dominance of AOA over AOB at higher temperatures. An instant temperature increase impacted the mesophilic AOB yield, corroborating the effect of maintenance requirement on production capacity. Model simulations of two realistic nitrification/denitrification plants were robust toward changing nitrifier yield in predicting effluent ammonium concentrations, whereas sludge composition was impacted. Summarized, a fast, precise and easily executable method was developed determining Ymax of ammonia and nitrite oxidizers in mixed communities.

Abstract: By 2050, the world population will have considerably expanded and the life standard of many will increase, yielding a 50% higher demand in protein (FAO, 2011), and even increases of 82 and 102% for diary and meat products, respectively (Boland et al., 2013). To provide in this increasing demand we are highly dependent on our classical fertilizer to food chain which has a high environmental impact and lacks efficiency. Nutrient losses cause eutrophication and biodiversity loss and the input of resources is already beyond the boundaries of environmental sustainability (Steffen et al., 2015). Phosphate fertilizers are made from phosphate rock (apatite), of which the reserves are predicted to be depleted within 50 100 years if we continue business as usual (Cordell et al., 2009). Next to problems related to the unbalanced geopolitical distribution with dominance in China and Morocco, the decreasing quality of the remaining apatite will result in an increasing environmental impact of fertilizer production. Finally, our traditional food production model requires 30% of all ice-free land, 70% of all available freshwater and produces up to one third of the global greenhouse gas emission, of which 80 to 86% is linked to agricultural production (Vermeulen et al., 2012). To ensure food security, nutrient recovery from waste streams can provide an important strategy. In this context, struvite ( ) crystallisation may be applied to recover phosphorus, along with some nitrogen. Reusing these nutrients as agricultural fertilizer on the field will lead to considerable losses to the environment. In contrast, their use to cultivate micro-organisms, e.g. for single cell protein (SCP), offers to potential of a near perfect conversion efficiency (Moed et al., 2015). At this moment, microalgae represent the most developed type of SCP, and are a promising protein source due to their growth rate, high nutritional quality and extremely high nutrient usage efficiency (Becker, 2007). Reliable solubilisation data are essential to design a technological strategy for struvite dosage in bioreactors for SCP production. The effect on solubility and solubilisation rate of relevant physicochemical parameters was studied experimentally in aqueous solutions. Because pH and temperature greatly affect solubilisation kinetics they were set at a constant value of 7 and 20°C respectively. The effect of some parameters on struvite solubility was already studied (Bhuiyan et al., 2007; Ariyanto et al., 2014; Roncal-Herrero and Oelkers, 2011), but solubilisation rates were not yet considered and pH was not controlled at a constant value. The chemical parameters considered in this study include the concentration of different common ions ( and ), foreign ions ( and the chelating agent ethylenediaminetetraacetic acid, EDTA) present in micro-algal cultivation media as well as ionic strength (as set by NaCl). The main physical parameter included was contact surface, through variation in initial particle size and as well as in struvite dosage concentration.

Abstract: The microalga Dunaliella salina has been widely studied for carotenogenesis, yet its protein production for human nutrition has rarely been reported. This study unveils the effects of growth phase and light regime on protein and essential amino acid (EAA) levels in D. salina. Cultivation under 24-h continuous light was compared to 12-h/12-h light/dark cycle. The essential amino acid index (EAAI) of D. salina showed accumulating trends up to 1.53 in the stationary phase, surpassing FAO/WHO standard for human nutrition. Light/dark conditions inferred a higher light-usage efficiency, yielding 597% higher protein and 1828% higher EAA mass on light energy throughout the growth, accompanied by 138% faster growth during the light phase of the light/dark cycle, compared to continuous light. The findings revealed D. salina to be especially suitable for high-quality protein production, particularly grown under light/dark conditions, with nitrogen limitation as possible trigger, and harvested in the stationary phase.

Abstract: High-rate activated sludge processes allow for the recovery of organics and energy from wastewaters. These systems are operated at a short sludge retention time and high sludge-specific loading rates, which results in a higher sludge yield and better digestibility than conventional, low-rate activated sludge. Little is known about the microbial ecology of high-rate systems. In this work, we address the need for a fundamental understanding of how high-rate microbial communities differ from low-rate communities. We investigated the high-rate and low-rate communities in a sewage treatment plant in relation to environmental and operational variables over a period of ten months. We demonstrated that (1) high-rate and low-rate communities are distinctly different in terms of richness, evenness and composition, (2) high-rate community dynamics are more variable and less shaped by deterministic factors compared to low-rate communities, (3) sub-communities of continuously core and transitional members are more shaped by deterministic factors than the continuously rare members, both in high-rate and low-rate communities, and (4) high-rate community members showed a co-occurrence pattern similar to that of low-rate community members, but were less likely to be correlated to environmental and operational variables. These findings provide a basis for further optimization of high-rate systems, in order to facilitate resource recovery from wastewater.

Abstract: High rate activated sludge (HRAS) processes have a high potential for carbon and energy recovery from sewage, yet they suffer frequently from poor settleability due to flocculation issues. The process of flocculation is generally optimized using jar tests. However, detailed jar hydrodynamics are often unknown, and average quantities are used, which can significantly differ from the local conditions. The presented work combined experimental and numerical data to investigate the impact of local hydrodynamics on HRAS flocculation for two different jar test configurations (i.e., radial vs. axial impellers at different impeller velocities) and compared the hydrodynamics in these jar tests to those in a representative section of a full scale reactor using computational fluid dynamics (CFD). The analysis showed that the flocculation performance was highly influenced by the impeller type and its speed. The axial impeller appeared to be more appropriate for floc formation over a range of impeller speeds as it produced a more homogeneous distribution of local velocity gradients compared to the radial impeller. In contrast, the radial impeller generated larger volumes (%) of high velocity gradients in which floc breakage may occur. Comparison to local velocity gradients in a full scale system showed that also here, high velocity gradients occurred in the region around the impeller, which might significantly hamper the HRAS flocculation process. As such, this study showed that a model based approach was necessary to translate lab scale results to full scale. These new insights can help improve future experimental setups and reactor design for improved HRAS flocculation.

Abstract: Purple non-sulfur bacteria (PNSB) show great potential for environmental and industrial biotechnology, producing microbial protein, biohydrogen, polyhydroxyalkanoates (PHAs), pigments, etc. When grown photoheterotrophically, the carbon source is typically more reduced than the PNSB biomass, which leads to a redox imbalance. To mitigate the excess of electrons, PNSB can exhibit several ‘electron sinking’ strategies, such as CO2 fixation, N2 fixation, and H2 and PHA production. The lack of a comprehensive (over)view of these redox strategies is hindering the implementation of PNSB for biotechnology applications. This review aims to present the state of the art of redox homeostasis in phototrophically grown PNSB, presenting known and theoretically expected strategies, and discussing them from stoichiometric, thermodynamic, metabolic, and economic points of view.

Abstract: Cost-efficient biological treatment of warm nitrogenous wastewaters requires the development of thermophilic nitrogen removal processes. Only one thermophilic nitrifying bioreactor was described so far, achieving 200 mg N L-1 d-1 after more than 300 days of enrichment from compost samples. From the practical point of view in which existing plants would be upgraded, however, a more time-efficient development strategy based on mesophilic nitrifying sludge is preferred. This study evaluated the adaptive capacities of mesophilic nitrifying sludge for two linear temperature increase patterns (non-oscillating vs. oscillating), two different slopes (0.25 vs. 0.08 °C d-1) and two different reactor types (floc vs. biofilm growth). The oscillating temperature pattern (0.25 °C d-1) and the moving bed biofilm reactor (0.08 °C d-1) could not reach nitrification at temperatures higher than 46°C. However, nitrification rates up to 800 mg N L-1 d-1 and 150 mg N g-1 volatile suspended solids d-1 were achieved at a temperature as high as 49°C by imposing the slowest linear temperature increase to floccular sludge. Microbial community analysis revealed that this successful transition was related with a shift in ammonium oxidizing archaea dominating ammonia oxidizing bacteria, while for nitrite oxidation Nitrospira spp. was constantly more abundant than Nitrobacter spp.. This observation was accompanied with an increase in observed sludge yield and a shift in maximal optimum temperature, determined with ex-situ temperature sensitivity measurements, predicting an upcoming reactor failure at higher temperature. Overall, this study achieved nitrification at 49°C within 150 days by gradual adaptation of mesophilic sludge, and showed that ex-situ temperature sensitivity screening can be used to monitor and steer the transition process.

Abstract: Human urine is a valuable resource for nutrient recovery, given its high levels of nitrogen, phosphorus and potassium, but the compositional complexity of urine presents a challenge for an energy-efficient concentration and refinery of nutrients. In this study, a pilot installation combining precipitation, nitrification and electrodialysis (ED), designed for one person equivalent (1.2 L-urine d(-l)), was continuously operated for similar to 7 months. First, NaOH addition yielded calcium and magnesium precipitation, preventing scaling in ED. Second, a moving bed biofilm reactor oxidized organics, preventing downstream biofouling, and yielded complete nitrification on diluted urine (20-40%, i.e. dilution factors 5 and 2.5) at an average loading rate of 215 mg N L-1 d(-1). Batch tests demonstrated the halotolerance of the nitrifying community, with nitrification rates not affected up to an electrical conductivity of 40 mS cm(-1) and gradually decreasing, yet ongoing, activity up to 96 mS cm(-1) at 18% of the maximum rate. Next-generation 16S rRNA gene amplicon sequencing revealed that switching from a synthetic influent to real urine induced a profound shift in microbial community and that the AOB community was dominated by halophilic species closely related to Nitrosomonas aestuarii and Nitrosomonas marina. Third, nitrate, phosphate and potassium in the filtered (0.1 mu m) bioreactor effluent were concentrated by factors 43, 2.6 and 4.6, respectively, with ED. Doubling the urine concentration from 20% to 40% further increased the ED recovery efficiency by similar to 10%. Batch experiments at pH 6, 7 and 8 indicated a more efficient phosphate transport to the concentrate at pH 7. The newly proposed three-stage strategy opens up opportunities for energy- and chemical-efficient nutrient recovery from urine. Precipitation and nitrification enabled the long-term continuous operation of ED on fresh urine requiring minimal maintenance, which has, to the best of our knowledge, never been achieved before. Abstract: Long-term human space exploration missions require environmental control and closed Life Support Systems (LSS) capable of producing and recycling resources, thus fulfilling all the essential metabolic needs for human survival in harsh space environments, both during travel and on orbital/planetary stations. This will become increasingly necessary as missions reach farther away from Earth, thereby limiting the technical and economic feasibility of resupplying resources from Earth. Further incorporation of biological elements into state-of-the-art (mostly abiotic) LSS, leading to bioregenerative LSS (BLSS), is needed for additional resource recovery, food production, and waste treatment solutions, and to enable more self-sustainable missions to the Moon and Mars. There is a whole suite of functions crucial to sustain human presence in Low Earth Orbit (LEO) and successful settlement on Moon or Mars such as environmental control, air regeneration, waste management, water supply, food production, cabin/habitat pressurization, radiation protection, energy supply, and means for transportation, communication, and recreation. In this paper, we focus on air, water and food production, and waste management, and address some aspects of radiation protection and recreation. We briefly discuss existing knowledge, highlight open gaps, and propose possible future experiments in the short-, medium-, and long-term to achieve the targets of crewed space exploration also leading to possible benefits on Earth.

Abstract: In een context van stijgende waterschaarste verkennen wij, naar ons weten voor het eerst in Vlaanderen, publieke steun voor de behandeling en het hergebruik van afvalwater als drinkwater. Vlaanderen is vandaag een van de weinige regio’s waar afvalwater reeds gerecycleerd wordt voor drinkwaterdoeleinden. Dit gebeurt op kleinschalig niveau en de uitbreiding hiervan is vandaag een van de Vlaamse beleidsdoelstellingen. Internationale voorbeelden toonden echter dat een gebrek aan publieke steun een aanzienlijk obstakel kan zijn. Vaak worden gezondheids- en veiligheidsbezorgdheden aangehaald als oorzaak voor het beperkte draagvlak. Minder is geweten over de socio-demografische distributie van dit draagvlak. Daarbovenop blijft er onduidelijkheid over de samenhang tussen socio-demografische kenmerken en gezondheids- en veiligheidsbezorgdheden. Met behulp van een enquête uitgevoerd in Vlaanderen (N=2309), bestudeerden wij ten eerste deze socio-demografische verschillen op basis van bivariate associaties (gender, opleidingsniveau, leeftijd en woonplaats). Ten tweede construeerden we een padmodel om te onderzoeken of deze verschillen verklaard kunnen worden aan de hand van gezondheids- en veiligheidsbezorgdheden. Onze resultaten toonden dat publieke steun voor afvalwaterhergebruik voor drinkwaterdoeleinden in Vlaanderen beperkt is. Het draagvlak was het laagst bij oudere mensen, vrouwen, lager geschoolde groepen en mensen die niet in de Provincie Antwerpen wonen. Voor een groot deel konden socio-demografische verschillen verklaard worden door hogere gezondheids- en veiligheidsbezorgdheden bij vrouwen, lager geschoolden en mensen uit West- en Oost-Vlaanderen. Dit suggereert een gebrek aan vertrouwen in waterexperts en -technologie bij bepaalde socio-demografische groepen, wat zich vertaalt in een verminderde publieke steun voor afvalwaterhergebruik. Op basis van deze bevindingen bespreken we een aantal potentiële actiestrategieën om publieke oppositie te anticiperen en proactief publieke steun te verwerven via doelgerichte (risico)communicatie.

Abstract: This study investigates the impact of lower temperature on short term and long term (down to 10 degrees C) on a completely anoxic anammox granular sludge process. This is the first time granular sludge Anammox is operated in pure anoxic condition in SBR and at low temperature. Conversion performance, kinetic parameters, sludge characteristics and microbial community were analyzed.

Abstract: Photoautohydrogenotrophic enrichments of wastewater treatment microbiomes were performed to obtain hypothetically high-potential specialist species for biotechnological applications. From these enrichment cultures, ten photoautohydrogenotrophic species were isolated: six Rhodopseudomonas species, three Rubrivivax members and Rhodobacter blasticus. The performance of these isolates was compared to three commonly studied, and originally photoheterotrophically enriched species (Rhodopseudomonas palustris, Rhodobacter capsulatus and Rhodobacter sphaeroides), designated as reference species. Repeated subcultivations were applied to improve the initial poor performance of the isolates (acclimation effect), which resulted in increases in both maximum growth rate and protein productivity. However, the maximum growth rate of the reference species remained 3–7 times higher compared to the isolates (0.42–0.84 d−1 at 28 °C), while protein productivities remained 1.5–1.7 times higher. This indicated that H2-based enrichment did not result in photoautohydrogenotrophic specialists, suggesting that the reference species are more suitable for intensified biomass and protein production. On the other hand, the isolates were able to provide equally high protein quality profiles as the references species, providing full dietary essential amino acid matches for human food. Lastly, the effect of metabolic carbon/electron switching (back and forth between auto- to heterotrophic conditions) initially boosted µmax when returning to photoautohydrogenotrophic conditions. However, the switch negatively impacted lag phase, protein productivities and pigment contents. In the case of protein productivity, the acquired acclimation was partially lost with decreases of up to 44 % and 40 % respectively for isolates and reference species. Finally, the three reference species, and specifically Rh. capsulatus, remained the most suitable candidate(s) for further biotechnological development.

Abstract: Nitrification and denitrification, the key biological processes for thermophilic nitrogen removal, have separately been established in bioreactors at 50 °C. A well-characterized set of kinetic parameters is essential to integrate these processes while safeguarding the autotrophs performing nitrification. Knowledge on thermophilic nitrifying kinetics is restricted to isolated or highly enriched batch cultures, which do not represent bioreactor conditions. This study characterized the stoichiometry and kinetics of two thermophilic (50 °C) nitrifying communities. The most abundant ammonia oxidizing archaea (AOA) were related to the Nitrososphaera genus, clustering relatively far from known species Nitrososphaera gargensis (95.5% 16S rRNA gene sequence identity). The most abundant nitrite oxidizing bacteria (NOB) were related to Nitrospira calida (97% 16S rRNA gene sequence identity). The nitrification biomass yield was 0.200.24 g VSS g−1 N, resulting mainly from a high AOA yield (0.160.20 g VSS g−1 N), which was reflected in a high AOA abundance in the community (5776%) compared to NOB (511%). Batch-wise determination of decay rates (AOA: 0.230.29 d−1; NOB: 0.320.43 d−1) rendered an overestimation compared to in situ estimations of overall decay rate (0.0260.078 d−1). Possibly, the inactivation rate rather than the actual decay rate was determined in batch experiments. Maximum growth rates of AOA and NOB were 0.120.15 d−1 and 0.130.33 d−1 respectively. NOB were susceptible to nitrite, opening up opportunities for shortcut nitrogen removal. However, NOB had a similar growth rate and oxygen affinity (0.150.55 mg O2 L−1) as AOA and were resilient towards free ammonia (IC50 > 16 mg NH3-N L−1). This might complicate NOB outselection using common practices to establish shortcut nitrogen removal (SRT control; aeration control; free ammonia shocks). Overall, the obtained insights can assist in integrating thermophilic conversions and facilitate single-sludge nitrification/denitrification.
