Abstract: At or below the surface of painted works of art, valuable information is present that provides insights into an object’s past, such as the artist’s technique and the creative process that was followed or its conservation history, but also on its current state of preservation. Typically, a (very) limited set of small paint samples is taken which provide direct access to the individual paint layers. The chemical build-up of these layers can then be investigated in great detail using various microscopic analytical methods. However, in recent years a new trend towards both elemental and chemical imaging techniques has been set which are capable of visualizing the (often) heterogeneous composition of painted objects on a macroscopic scale. In this dissertation, various forms of specificity attainable with X‑ray powder diffraction (XRPD) imaging are explored: at the chemical, material and spatial level. This high specificity is illustrated throughout several applications stemming from the field of cultural heritage, both at the macroscopic (MA) and microscopic (µ) scale. As a first step, XRPD imaging was transformed to a transportable instrument that can be employed for the in situ investigation of artworks, e.g., inside museums and conservation workshops. With this unique instrument large‑scale maps (cm2 – dm2) reflecting the distribution of crystalline phases on/below the surface of flat painted artefacts can be visualized in a noninvasive manner. In this way compound-specific information was attained which can be related to original pigments or materials that have been added in a later stage and even degradation/secondary products that have formed spontaneously inside the paint layers. Additionally, with MA‑XRPD imaging it was possible to link quantitative information of pigment compositions and preferred orientation effects to the 2D compound‑specific distribution images, allowing for a further distinction between very similar artists’ materials. Furthermore, promising results for the limited depth-selectivity of this technique, obtained by exploiting the small shift in the position of the diffraction signals originating from the layered sequence of the pigments, are shown. Finally, a minute paint sample from Wheat stack under a cloudy sky by Van Gogh was investigated at a synchrotron radiation facility with tomographic µ‑XRPD imaging at the microscopic scale. The high chemical and spatial specificity of this imaging method was exploited to further elucidate the degradation pathway of the red lead pigment.

Abstract: Nowadays the electronic structure calculations allow the study of complex systems determining the hyperfine parameters measured at a probe atom, including the presence of crystalline defects. The hyperfine electric parameters have been measured by Mossbauer spectroscopy in silicon materials implanted with Mn-57 ->,Fe-57 ions, observing four main contributions to the spectra. Nevertheless, some ambiguities still remain in the Fe-57 Mossbauer spectra interpretation in this case, regarding the damage configurations and its evolution with annealing. In the present work several implantation environments are evaluated and the Fe-57 hyperfine parameters are calculated. The observed correlation among the studied local environments and the experimental observations is presented, and a tentative microscopic description of the behavior and thermal evolution of the characteristic defects local environments of the probe atoms concerning the location of vacancies and interstitial Si in the neighborhood of Fe-57 ions in substitutional and interstitial sites is proposed. Abstract: The formation and distribution of metal soaps produced as a result of the reactivity and aging of the materials in a fifteenth-century egg tempera and oil paintings on wood are presented. The painting technique involves the application of several paint layers over a ground using, sometimes in the same paint layer sequence, drying oil and egg yolk binders. We show, with a selection of examples, how the use of thin sections and a combination of various micro-sensitive analytical techniques is adequate to obtain the high-quality data necessary for the unambiguous identification of metal soaps and metal oxalates as well as their distribution in the paint layers. The techniques include micro infrared spectroscopy (μSR-FTIR) and micro X-ray diffraction (μSR-XRD) with synchrotron radiation, optical microscopy (OM), and scanning electron microscopy with energy dispersive X-ray spectroscopy (SEM-EDS). The data obtained sheds light about the underlying reaction and aging mechanisms happening in each paint layer and among them. This helps to define the state of conservation of the artworks.

Abstract: Aerosol samples have been collected over the southern bight of the North Sea from an aircraft. In this way, 96 samples were taken for single-particle analysis during 16 flights. Almost 45 000 individual particles were analyzed with electron probe X-ray microanalysis. More than 5000 of these were found to contain significant concentrations of one or more of the heavy metals Cr, Pb, and Zn. With the help of hierarchical, nonhierarchical, and fuzzy clustering techniques, various heavy metal-containing particle types could be identified. Significant differences in abundances were detected in the North Sea heavy metal aerosol, depending on the origin of the air masses. In samples with continental influence 50 times more Zn- and Pb-containing particles were found than in samples with a marine history. For Cr, on the other hand, we found abundances in the marine sector that were one-third of the values for continental sectors. This might point to a rather undefined marine source, which could be the recycling of previously deposited material by reinjection into the atmosphere by sea spray. The highest values for Cr-, Pb-, and Zn-containing particles were always detected under southeastern wind directions.

Abstract: Laser microprobe mass spectrometry is used to identify intrarenal microliths; they appear to consist of either oxalate, urate or phosphate. Crystalline and amorphous deposits in rat and human kidney are pin-pointed by the laser beam and their chemical composition determined by mass spectrometry. The method has the potential for wide application in the identification of single organic, inorganic or combination crystals in histological sections.

Abstract: This paper presents a previously unexplored biosensing strategy for detecting gliadin which exploits the crosslinking of gliadin with collagen, catalyzed by transglutaminase at the interfacial electron transfer rate, on a modified electrode. The process is monitored by electrochemical impedance spectroscopy using a glassy carbon electrode coated with a collagen layer. To validate the specificity of the response as well as to eliminate possible interferences from other proteins, such as soy protein or casein, the captured gliadin is further reacted with a specific anti-gliadin antibody. Changes in charge transfer resistance, measured from the Nyquist plots, scale linearly with the gliadin concentration in the range 5-20 mg/L, a range suitable for testing the gliadin concentration in gluten-free food commodities.

Abstract: It is generally known that radiographic inspection of 1517th century paintings can easily be done with a polychromatic X-ray source using a voltage between 20 kV and 40 kV in combination with classic X-ray films. Unfortunately, the spatial structure of numerous 19th and early 20th century paintings cannot be visualized with conventional radiography due to several reasons such as the use of lead white grounds or low absorbing pigments. Radiographic images are blurred or worse, they do not contain the picture of the painting. During the last decades, many technological innovations have been introduced in the field of radiography but their possibilities in cultural heritage have not been explored in full detail. In our investigation we used phosphor imaging plates, energy dispersive detectors and CCD-cameras in combination with synchrotron radiation and conventional X-ray tubes in order to improve the quality of radiographic images. Several promising techniques that could improve the quality of radiographs of paintings were identified.

Abstract: Erythrocyte selenium-dependent glutathione peroxidase activity was measured in psoriatic Danes, before and after their four-week balneological therapy at the Ein-Bokek International Psoriasis Treatment Center, on the Dead-Sea shore in Israel. The drinking water in Ein-Bokek was found to be rich in selenium, a trace element with anticarcinogenic properties and of great importance in human nutrition and health. The most reliable biological parameter for increase in selenium bioavailability is the erythrocytes' glutathione-peroxidase activity. As psoriasis is a proliferative skin disease, the activity of this enzyme was assayed in 35 psoriatic Danes and in 25 long-term local hotel workers, as well as in 34 volunteers drinking low-selenium water. The glutathione peroxidase activity in the psoriatic patients increased significantly during their four-week stay in Ein-Bokek. Erythrocyte glutathione peroxidase activity in the hotel workers was 50% higher than that in the healthy volunteers consuming low-selenium water. A possible role of selenium in psoriasis is suggested.

Abstract: Iron (Fe) and other trace elements such as Zn, Mn, Ni and Cu are known as key-factors in marine biogeochemical cycles. It is believed that ocean primary productivity blooms in iron deficient regions can be triggered by iron in aeolian dust. Up to now, scarce aerosol elemental composition, based on measurements over sea at the Western South Atlantic (WSA), exist. An association between the Patagonian semi-desert dust/Fe and chlorophyll-a variability at the Argentinean continental shelf is essentially inferred from models. We present here experimental data of Fe enriched aerosols over the WSA between latitudes 22°S62°S, during 4 oceanographic campaigns between 2002 and 2005. These data allowed inferring the atmospheric Fe flux onto different latitudinal bands which varied from 30.4 to 1688 nmolFe m−2 day−1 (October 29thNovember 15th, 2003); 5.831586 nmolFe m−2 day−1 (February 15thMarch 6th, 2004) and 4.73586 nmolFe m−2 day−1(October 21stNovember 5th, 2005).
