Abstract |
Dit onderzoek presenteert een overzicht van de mogelijkheden van LA-ICP-MS als een techniek voor kwantitatieve analyse, bepalen van isotoopverhoudingen en elementaire beeldvorming. Hiervoor wordt de techniek zelf voorgesteld en worden de belangrijkste parameters die een invloed hebben op de kwaliteit van een LA-ICP-MS meting beschreven. Voor kwantitieve analyse worden de huidige calibratiestrategieën voor LA-ICP-MS toegelicht, zowel op het vlak van selectie en bereiding van standaarden als op het vlak van dataverwerking. Daarnaast werd een nieuwe calibratiestrategie voorgesteld, gebaseerd op een lineaire combinatie van standaarden. Een case study met de traditionele calibratiestrategie uit de literatuur werd uitgevoerd en vergeleken met data van SEM-EDX om een referentiekader te schetsen voor de performantie van de techniek. In een tweede case study werd onze nieuwe calibratiestrategie toegepast en werd er uitsluitend gebruik gemaakt van LA-ICP-MS, wat uitstekende resultaten opleverde. Met het oog op het bepalen van isotoopverhoudingen, werd er onderzocht wat de limieten zijn van het gebruik van een lage resolutie quadrupool massaspectrometer voor deze bepaling. Dit stelde ons in staat om de te verwachten accuraatheid en precisie in te schatten. De performantie bleek goed genoeg te zijn om weapon-grade plutonium te onderscheiden van global fall-out, zoals het werd aangetoond in de case study omtrent dit onderwerp. Uiteindelijk hebben we ook LA-ICP-MS geëvalueerd als een techniek voor elementaire beeldvorming, waarbij we tevens de meest courante multivariate data analyse hebben beschreven. Om de techniek te plaatsen tegenover andere elementaire technieken, werd een historisch verffragment geanalyseerd dat in het verleden reeds geanalyseerd was met SEM-EDX and μ-XRD. Op die manier konden we de resultaten accuraat vergelijken met de andere technieken. |

Abstract |
Triammonium aurin tricarboxylate (aluminon) has been used to localize aluminum in 2 μm sections of undecalcified, methyl methacrylate embedded bone obtained from patients with terminal chronic renal failure. Aluminum appeared in four cases as bright red lines at the mineralized-bone boundary. In two cases, however, purplish lines were found and one patient showed red as well as purplish lines. Laser microprobe mass analysis (LAMMA) identified aluminum at the location of the red lines and both aluminum and iron at the purplish lines. Furthermore, both iron and aluminum were found in histiocytic bone marrow cells, which showed brownish aluminon staining. It appears that when aluminum and iron occur together, aluminon staining may yield aberrant results. This study shows that LAMMA can be used for the identification of elements sought by histochemical methods and thus permits the evaluation of their staining effects. |

Abstract |
Individual Antarctic aerosol particles in the 0.54 μm aerodynamic diameter range were analyzed using laser microprobe mass analysis (LAMMA). As they were sampled near the ocean, the great majority consists of seasalt, transformed to various degrees in the atmosphere. Major alterations include the association of an excess sulfate and methane sulfonate with these particles. Sulfate-rich particles containing little or no chloride were found mostly in the smallest size fraction (0.51 μm), where they account for some 5% of all particles: they are most likely highly transformed seasalt. Aluminosilicates, on the other hand, only appear among the coarser particles: they represent 2% of the particulates in the 24 μm range. The remainder of the aerosol consists of organic, Fe-rich, K-rich and Zn-rich particles. The latter groups have very low abundances: always less than 1% of the population of the impactor stage(s) onto which they were collected. |

Abstract |
By means of laser microprobe mass analysis (LAMMA) platinum was detected in the renal proximal tubular cells of a dog that had been intravenously adminstered the antitumor drug cisplatin (5 mg per kg body weight). No definite subcellular localization of the heavy metal was obtained. Sample prparation and analytical features are examined to increase spatial resolution of analysis while maintaining sufficient detection efficiency. The LAMMA method is destructive, but the amount and type of evaporated material can readily be determined when using LAMMA in combination with transmission electron microscopy. Instrumental optimization and standardization of mass signals is possible by using platinum-loaded, ion chelating resin beads embedded and sectioned with the tissue. |

Abstract |
Laurdan is one of the most used fluorescent probes for lipid membrane phase recognition. Despite its wide use for optical techniques and its versatility as a solvatochromic probe, little is known regarding its use as molecular rotor, for which clear evidence is found in the current study. Although recent computational and experimental studies suggest the existence of two stable conformations of laurdan in different membrane phases, it is difficult to experimentally probe their prevalence. By means of multiscale computational approaches, we prove now that this information can be obtained through the optical properties of the two conformers, ranging from one-photon absorption over two-photon absorption to the first hyperpolarizability. Fluorescence decay and anisotropy analyses are performed as well and stress the importance of laurdan's conformational versatility. As a molecular rotor and with reference to the distinct properties of its conformers, laurdan can be used to probe biochemical processes that change the lipid orders in cell membranes. |

Abstract |
A study has been conducted on the leaching of two fungicides, tebuconazole and cymoxanil, in a soil amended with spent mushroom substrate (SMS), with an evaluation of how different factors influence this process.The objective was based on the potential use of SMS as a biosorbent for immobilizing pesticides in vulnerable soils, and the need to know how it could affect the subsequent transport of these retained compounds. Breakthrough curves (BTCs) for C-14-fungicides, non-incubated and incubated over 30 days, were obtained in columns packed with an unamended soil (S), and this soil amended with SMS at rates of 5% (S + SMS5) and 50% (S + SMS50) under saturated and saturated-unsaturated flows. The highest leaching of tebuconazole (> 50% of the total C-14 added) was found in S when a saturated water flow was applied to the column, but the percentage of leached fungidde decreased when a saturated-unsaturated flow was applied in both SMS-amended soils. Also a significant decrease in teaching was observed for tebuconazole after incubation in the column, especially in S + SMS50 when both flows were applied. Furthermore, cymoxanil leaching was complete in S and S + SMS when a saturated flow was applied, and maximum peak concentrations were reached at 1 pore volume (PV), although BTCs showed peaks with lower concentrations in S + SMS. The amounts of cymoxanil retained only increased in S + SMS when a saturated-unsaturated flow was applied. A more relevant effect of SMS for reducing the leaching of fungidde was observed when cymoxanil was previously incubated in the column, although mineralization was enhanced in this case. These results are of interest for extending SMS application on the control of the leaching of fungicides with different physicochemical characteristics after different ageing times in the soil and water flow conditions applied. Abstract |
In air particulate matter, sampled at a suburban site near the city of Antwerp, Belgium, more than 1500 individual Pb-containing aerosol particles with projected diameters between 0.2 and 15 μm were analyzed by automated electron probe X-ray micro analysis (EPXMA) for 26 elements and morphological features. Laser microprobe mass analysis (LAMMA) was used to detect elements with Z < 11, particularly ammonium compounds. The Pb-containing particles were classified into five main classes: Pb-sulfates, Pb-halides, soil related Pb, Pb associated with medium atomic number elements and Pb associated with high atomic number elements. Each class was divided into several distinct particle types, of which the abundance (in number and mass %), the mean projected particle diameter, and the chemical composition were determined. Auto exhaust products are responsible for Pb-sulfates and Pb-halides, making up respectively 66.7 and 27.8% by mass of the Pb-containing particles. Ammonium sulfate coatings were found to be present on nearly all Pb particles. The observations were correlated with daily concentrations of Pb and Br, obtained by particle induced X-ray emission analysis (PIXE). |

Abstract |
According to Thompson’s principle of similarity, the area of an object should be proportional to its length squared. However, leaf area–length data of some plants have been demonstrated not to follow the principle of similarity. We explore the reasons why the leaf area–length allometry deviates from the principle of similarity and examine whether there is a general model describing the relationship among leaf area, width and length. We sampled more than 11,800 leaves from six classes of woody and herbaceous plants and tested the leaf area–length allometry. We compared six mathematical models based on root-mean-square error as the measure of goodness-of-fit. The best supported model described a proportional relationship between leaf area and the product of leaf width and length (i.e., the Montgomery model). We found that the extent to which the leaf area–length allometry deviates from the principle of similarity depends upon the extent of variation of the ratio of leaf width to length. Estimates of the parameter of the Montgomery model ranged between 1/2, which corresponds to a triangular leaf with leaf length as its height and leaf width as its base, and π/4, which corresponds to an elliptical leaf with leaf length as its major axis and leaf width as its minor axis, for the six classes of plants. The narrow range in practice of the Montgomery parameter implies an evolutionary stability for the leaf area of large-leaved plants despite the fact that leaf shapes of these plants are rather different. |

Abstract |
The microalga Dunaliella salina has been widely studied for carotenogenesis, yet its protein production for human nutrition has rarely been reported. This study unveils the effects of growth phase and light regime on protein and essential amino acid (EAA) levels in D. salina. Cultivation under 24-h continuous light was compared to 12-h/12-h light/dark cycle. The essential amino acid index (EAAI) of D. salina showed accumulating trends up to 1.53 in the stationary phase, surpassing FAO/WHO standard for human nutrition. Light/dark conditions inferred a higher light-usage efficiency, yielding 597% higher protein and 1828% higher EAA mass on light energy throughout the growth, accompanied by 138% faster growth during the light phase of the light/dark cycle, compared to continuous light. The findings revealed D. salina to be especially suitable for high-quality protein production, particularly grown under light/dark conditions, with nitrogen limitation as possible trigger, and harvested in the stationary phase. |

Abstract |
Wastewater is typically treated by the conventional activated sludge process, which suffers from an inefficient overall energy balance. The high-rate contact stabilization (HiCS) has been proposed as a promising primary treatment technology with which to maximize redirection of organics to sludge for subsequent energy recovery. It utilizes a feast famine cycle to select for bioflocculation, intracellular storage, or both. We optimized the HiCS process for organics recovery and characterized different biological pathways of organics removal and recovery. A total of eight HiCS reactors were operated at 15 degrees C at short solids retention times (SRT; 0.24-2.8 days), hydraulic contact times (t(c); 8 and 15 min), and stabilization times (t(s); 15 and 40 min). At an optimal SRT between 0.5 and 1.3 days and t(c) of 15 min and t(s) of 40 min, the HiCS system oxidized only 10% of influent chemical oxygen demand (COD) and recovered up to 55% of incoming organic matter into sludge. Storage played a minor role in the overall COD removal, which was likely dominated by aerobic biomass growth, bioflocculation onto extracellular polymeric substances, and settling. The HiCS process recovers enough organics to potentially produce 28 kWh of electricity per population equivalent per year by anaerobic digestion and electricity generation. This inspires new possibilities for energy-neutral wastewater treatment. |

Abstract |
Trace elements can influence dental health, possibly by altering tooth resistance during preeruptive development. Therefore, it was investigated whether lead and fluoride would be incorporated into the calcifying matrices or the cellular parts of tooth germs in vitro. Using laser microprobe mass analysis, the localization of lead and fluoride was studied in the different layers or tooth germs that had been cultured in a medium to which PbCl2 of NaF had been added in different concentrations. Both elements could only be detected in the dentine layer. Hence, the enamel organ in the secretory stage of tooth development excludes lead and fluoride from the enamel, even when enamel formation by the ameloblasts is visibly disturbed. Furthermore, there seemed to be a process of saturation in the accumulation of lead and fluoride in the dentine. |
